Objective To investigate the research status, hotspots, and trends in PIK3CA gene mutations in metastatic colorectal cancer (mCRC).
Method Relevant literature on PIK3CA gene mutations in mCRC was retrieved from the Web of Science Core Collection, covering the period from database inception to July 17, 2025. CiteSpace (version 6.3.R1) was employed to conduct visual analyses of annual publication volume, citation counts, countries, institutions, and keywords. Additionally, keyword co-occurrence, a clustering and burst analyses were carried out.
Results A total of 514 publications were included. The annual publication volume exhibited a generally stable trend over time. The University of Texas System was the most productive institution. The United States ranked first in both publication count and centrality. Currently, research hotspots include the mechanistic role of PIK3CA in mCRC, its involvement in therapy and resistance, and the application of liquid biopsy technologies.
Conclusion The PIK3CA gene holds significant therapeutic potential in mCRC. Further investigation into PIK3CA-related molecular mechanisms and the development of combinatorial treatment strategies may offer novel approaches for the clinical management of this disease.
